Medicine. For my PhD, I studied electrical synapse-mediated oscillations in the membrane potential of neurons under John Welsh. For my postdoc, I studied stem cell biology in Lorenz Studer and Viviane Tabar’s labs at Memorial Sloan-Kettering. I also trained in neurosurgery at Cornell and Memorial Sloan-Kettering. At NYU, I am a neurosurgeon-scientist focusing my research and clinical practice on brain tumors, and glioma in particular. Our lab is interested in how gliomas form and progress. Lab projects range from understanding the function of novel cell surface receptors relevant to glioma biology; to how changes in chromatin folding may predispose normal neural progenitors to transform into glioma cells. My main motivation is to make a difference for my patients, with whom I form strong bonds. I care deeply about them and know that doing good surgery is not enough; I therefore try to understand the biology of brain tumors to really make a difference in their care. Like all scientists, I am addicted to the thrill of scientific discovery, but also love doing surgery to remove brain tumors. Outside of work, I love music and dance; and am passionate about helping students and colleagues reach their full potential and flourish. recently described that GPR133, an orphan member of the adhesion family of G protein-coupled receptors (GPCR), is necessary for glioblastoma growth. GPR133 is expressed in all glioblastomas, regardless of molecular subtype, but is absent from normal brain tissue. This expression pattern suggests a favorable therapeutic profile. We are currently elucidating basic mechanisms of action of GPR133, and other adhesion GPCRs, such as CD97, but also developing them translationally as novel therapeutic targets. https://images.squarespace-cdn.com/content/v1/5aea234b2714e5e878480357/1636564241034-JTT6B3NLQTO52BGQP3NQ/human+neurogenesis+figure.png Research Projects - Developmentally regulated 3D chromatin architecture and enhancer activation in human neurogenesis We discovered that transcription of SOX2, which encodes a transcription factor critical to neural progenitor self-renewal and multipotency, is regulated by a distal enhancer through 3D chromatin folding. Both the enhancer activation and the chromatin looping occur specifically in neural progenitors but not embryonic stem cells or other germ layers. We believe that these waves are relevant to tumor growth. A new and and exciting project in the lab focuses on understanding the mechanisms underlying these waves, their importance to tumor biology, and the signaling mechanisms initiated by calcium flux in glioma. Over the past decade, several lines of evidence have indicated that not every cell in these tumors is equal. Another way to state this is that some tumor cells in GBM are more important than the rest. In biological terms, this inequality translates into a cellular hierarchy, whose apex is occupied by cancer stem cells. Tissue homeostasis is maintained by stem cells that generate defined lineages of specialized differentiated cells. Not surprisingly, GBM tumors are remarkably complex at the histologic level, which begs the question: Does a single cell type with stem-like properties produce the entire spectrum of tumor lineages and histologies or is the cancer stem cell population heterogeneous? Using human GBM cultures, we recently discovered that within any given GBM tumor there are multiple tumor cell types that fulfill stem cell criteria. Great mechanistic insights and a terrific translational aspect. Niklas showed that: 1) CD97 is an adhesion GPCR that is de novo expressed in GBM compared to healthy brain; 2) CD97 promotes tumorigenesis and glycolytic metabolism in GBM via MAPK signaling; 3) Phosphorylation of the C terminus of CD97 leads to β-arrestin recruitment; and 4) GBM cells can be selectively killed using a CD97-targeting antibody-drug conjugate. PTK7 binding produces positive allosteric effects on GPR133 signaling when the two proteins interact in trans (on neighboring cells).
